Microbial phenolic metabolites 3-(3′,4′-dihydroxyphenyl)propanoic acid and 3′,4′-dihydroxyphenylacetic acid prevent obesity in mice fed high-fat diet

Obesity is associated with numerous metabolic disorders, and dietary polyphenols have been confirmed to beneficial effects on the metabolism in obesity. However, effect of 3-(3′,4′-dihydroxyphenyl)propanoic acid (DHPA) 3′,4′-dihydroxyphenylacetic (DHAA), two main metabolites polyphenols, obesity remains poorly understood. In this study, DHPA DHAA were found alleviate obesity, as well regulate insulin resistance, lipid metabolism, oxidative stress response high-fat diet (HFD) mice. Surprisingly, 16S rRNA sequencing UHPLC-Q-TOF/MS demonstrated that only slightly disturbed intestinal microbiome, but significantly altered urine metabolome HFD mice mainly by regulating pentose glucuronate interconversion, tyrosine phosphate TCA cycle indicated KEGG analysis. Correlation analysis revealed differential are strongly body weight, blood glucose, level, SOD enzyme activity. Our results exert their anti-obesity important pathways.
